What is Retatrutide?
Retatrutide is a single-chain synthetic peptide developed as an agonist at three receptors: glucose-dependent insulinotropic polypeptide (GIP), glucagon-like peptide-1 (GLP-1) and glucagon (GCG). It is built on a GIP-based peptide backbone carrying a fatty diacid moiety that promotes albumin binding and extends its half-life.
Phase 2 clinical results were published in 2023 and the compound is the subject of ongoing phase 3 programmes, which gives it a fast-growing literature. In the laboratory it is used to study multi-receptor incretin and glucagon signalling, energy expenditure and hepatic lipid metabolism, and as a comparator for dual and single receptor agonists.
